In Vitro Cytostatic Effect of Extract from a Marine Streptomyces sp. on Plasmodium Falciparum 3D7

چکیده
This study was designed to examine the stage dependency of growth inhibition by Streptomyces sp. crude extract and to determine whether its antimalarial activity is cytostatic or cytocidal. Growth inhibition was assessed by differences in growth appearance between treated and control parasites. Using synchronized In vitro cultures of Plasmodium falciparum strain 3D7, growth inhibition was detected within a single parasite cycle. Ring forms and trophozoite stages were sensitive to the inhibitory effect of the crude extract but antimalarial activity was suggested by evidence of late developmental stage. However, evidence of subsequent recovery, occurred when the treated cells were washed with a new medium, and then incubated for 24 hours. At this stage ring forms and the trophozoites showed a successful development into late stages. These observations suggest that the Streptomyces sp. crude extract may have a cytostatic effect on Pl. falciparum.

Background and Objective: Malaria is one of the most life-threatening infectious diseases worldwide. Transmission of the parasite from human to vector mosquitoes is carried out by the gametocyte of the Plasmodium parasite, while these cells are not involved in the symptoms of the disease. The control of the human to mosquito transmission stage of the parasite life cycle by antigametocyte drugs...

Mass-directed isolation of the CH(2)Cl(2)/CH(3)OH extract from a marine sponge of the genus Pseudoceratina resulted in the purification of a new antimalarial bromotyrosine alkaloid, psammaplysin H (1), along with the previously isolated analogs psammaplysins G (2) and F (3). The structure of 1 was elucidated following 1D and 2D NMR, and MS data analysis. All compounds were tested in vitro again...

Some Dorema species are used in Persian traditional medicine. In the present study the total extract from the roots of Dorema hyrcanum Koso-Pol. was investigated for its in vitro (pLDH assay) and in vivo (Peters’ 4-days suppressive test) antiplasmodial effects and assessed for cytotoxicity against the normal cell line MDBK (MTT test). The IC50 values for a chloroquine- sensitive (3D7) and a chl...
